Day after day and month after month, research never stops, and my thinking never stops. We urgently need to strengthen basic research, but what are the key points in basic research? To do a good job, one must first make use of their tools. Basic research needs to make breakthroughs, and to seize the initiative in field development, key research and technological innovation must take a step forward. This is because innovation in research technology often determines whether one can take the lead and make important breakthroughs and discoveries in relevant fields. Let's talk about the paleogenetics that I am engaged in! It is a cutting-edge interdisciplinary field between Earth and life sciences, and key research techniques play a crucial role. My team and I have stagnated in the exploration of many key scientific issues because of the bottleneck of obtaining key technologies of ancient DNA. What should we do? Should we wait for others to conquer this technology before continuing our own research? If that were the case, we would definitely lose the opportunity. Never wait! There is no "sharp edge" in scientific research, we make it ourselves and grind it ourselves. After years of joint efforts, we have caught up in technology! By combining ancient DNA with high-throughput sequencing technology, we have established a new single strand library preparation technology to obtain high-quality genomes of extinct ancient humans, and found that these ancient genes of extinct ancient humans have an important impact on modern humans in terms of physiological function, disease immunity and environmental adaptation; We have developed the ancient nuclear genome capture technology to obtain the oldest modern human genome in East Asia and the genomes of populations in southern China around ten thousand years ago. We have answered important scientific questions about the evolution characteristics of populations in East Asia, South and North Asia, and the origin of the Austronesian language family, filling an important gap in the world's human evolution history. These technologies have greatly expanded the sample range that can be used for ancient DNA research, and will strongly promote the development of anthropology, evolutionary genetics, population genetics and other related disciplines.
