AI Model Design Six Proteins with Better Performance

Artificial intelligence (AI) protein design is moving towards "faster, better, stronger". The team from the Brigham College of Massachusetts General Hospital and the Beth Israel Deaconess Medical Center has developed an AI tool called EVOLVEpro, which is considered a major breakthrough in the field of protein engineering. The team presented six proteins with different applications designed using this tool in the latest issue of Science magazine, demonstrating that EVOLVEpro can improve protein stability, accuracy, and efficiency. The concept of protein engineering has a long history, but with the rise of AI and large-scale language models, this field is undergoing profound changes. The protein language model can understand the "grammar" of proteins and provide specific suggestions for improving proteins by analyzing protein sequences in vast genomic databases. Similar to the latest large-scale language models, EVOLVEpro serves as an additional layer of functionality that enables in-depth reasoning and thinking before responding. The team designed six proteins using EVOLVEpro. The results showed that the adhesion of the two monoclonal antibodies optimized by EVOLVEpro to the target was enhanced by 30 times; The efficiency of gene editing performed by micro CRISPR nucleases has increased by 5 times; The protein used for gene editing has doubled its ability to insert sequences into different positions of the genome; The efficiency of Bxb1 integrase in implanting DNA fragments into cells for programmable gene integration has increased fourfold; The T7 RNA polymerase used for RNA synthesis has increased its ability to accurately replicate RNA by 100 times. The team pointed out that the biggest advantage of this tool is that it is not limited by natural evolution. With the help of AI, they can optimize proteins based on specific needs, creating proteins with better performance, faster speed, and higher strength, making them more effective in binding to targets, thereby improving treatment methods or enhancing their functionality. This technology means that humans will not only be able to design proteins that are comparable to natural products, but also have the ability to tackle challenges that nature has never encountered before, including developing more precise antibodies to treat autoimmune diseases or cancer, developing more effective virus vaccines, or cultivating crops with higher nutritional value or stronger carbon dioxide absorption capacity.
