The University of Hong Kong has developed the world's first platform for screening precise gene editing tools

On the 19th, the University of Hong Kong School of Medicine announced that a research team has developed the world's first platform for screening precise gene editing tools, and has now submitted a patent application for this technology. It is reported that this research was led by Huang Zhaolin, an associate professor of the School of Biomedical Sciences of the HKU School of Medicine, and was funded by the Hong Kong Research Grants Council, the National Natural Science Foundation for Distinguished Young Scholars, and the InnoHK Research Center for Cancer and Immunology, the Innovation and Technology Commission of the Hong Kong Special Administrative Region Government. The Medical College of Hong Kong University introduced that the new testing platform developed by the research team can simultaneously test and modify hundreds of precise base editor variants, breaking through the limitations of current gene editing tools that need to be tested one by one, and quickly identifying the most suitable and effective gene therapy tools. To improve the efficiency of the existing base editor system, the team further expanded the application of the platform to modify the stem ring 2 region of the single stranded guide RNA skeleton, and screened in the base editor system. Two new skeleton variants, SV48 and SV240, were successfully identified, which were superior to the native type, achieving higher (up to 2.2 times) base editing efficiency. In addition, the team has demonstrated that the platform can not only recognize and screen base editors, but also be compatible with other precise gene editor systems, such as lead editors. Huang Zhaolin, associate professor at the School of Biomedical Sciences at the University of Hong Kong, stated that the new platform has powerful functions and effectively accelerates the development of a new generation of precise gene editors and their application in future treatments. Huang Zhaolin used the quick checkout channel in the store as an analogy to explain that "due to the special markings on all goods (i.e. base editor variants), when using a self-service checkout machine to scan, we only need to put all goods into the basket of the checkout machine once, and the scanner can automatically identify all goods and complete payment (i.e., perform base editor performance analysis in the case) .
