Progress in anti-aging targeted drug research: only remove aging cells, without harming "innocent"

The joint team of Vision China has developed a new Senolytics prodrug, KSL0608-Se, which can play its role through light induction after accurately anchoring aging cells by integrating near-infrared (NIR) fluorescence labeled enzyme substrate navigation, bio-orthogonal targeted anchoring, photodynamic therapy and other technologies, and has realized the precise identification and clearance of aging cells by single-cell resolution. With the development and progress of society, modern people have a longer life span, but they are often troubled by diseases when they go through the process of aging. Through research, scientists have found that one of the important pathogenic mechanisms is that the chronically accumulated aging cells in the elderly can not be removed in a timely and effective manner. On March 12, the reporter of Science and Technology Daily learned that Professor Li Jian of East China University of Science and Technology and Professor Guo Yuan of Northwestern University jointly proposed and verified a new drug technology strategy to eliminate aging cells, which can be used to selectively remove aging cells and slow down the aging of mice's functional organs. The relevant research papers were published in the journal Nature Aging. "Longevity medicine" faces some problems. The cells in the body are constantly metabolized, such as liver cells are updated every 300-500 days, platelets are updated every 7-14 days, and epidermal cells are updated almost every day. Normally, these aging cells will be cleaned up by the immune system. However, due to aging and other reasons, the function of the immune system will gradually decline. Once these aging cells cannot be cleaned up, they will accumulate in the body. Some aging cells with abnormal functions also attack healthy cells, known as "zombie cells". The aging cells with abnormal functions will send out a series of potentially harmful chemical signals to "encourage" nearby cells to enter the same aging state. These signals include pro-inflammatory cytokines, chemokines and extracellular matrix proteinases, which together form the senescence-related secretory phenotype (hereinafter referred to as SASP). The existence of SASP can lead to many problems, such as reducing tissue function, increasing the level of chronic inflammation, and ultimately increasing the risk of cancer. So can we treat age-related diseases or even reverse aging by eliminating or improving "zombie cells"? According to this idea, scientists have developed therapeutic drugs targeting "zombie cells", which are mainly divided into two categories: one is called "Senolytics", which is used to selectively eliminate aging cells; The other class is called "Senomology", which is used to prevent the harmful cell external effects of aging cells. But people still face many problems when they want to find a "longevity medicine" that can really solve aging. Senomorphics usually needs to be administered continuously for a long time to "cure the symptoms but not the root cause". The effectiveness of Senolytics has been confirmed in mouse experiments. After selectively clearing "zombie cells", scientists have successfully extended the life span of mice and prevented or terminated senile diseases. Therefore, people have high hopes for Senolytics, which directly targets aging cells. At present, many preclinical evidences have proved that Senolytics can selectively clear aging cells and interfere with various aging
